南27線以[[將軍區]]長榮里南側的[[南19線]]忠興路叉路為起點，由此往西南略行一段路程後越過區界來到[[佳里區]]漳洲里的範圍，在「後港腳」{{r|origin1}}東側交會東西向的[[南28線]]，再經過該里內最大聚落─「番子寮」{{r|origin2}}。本線沿番子寮西側朝西南向前行時，在延平國小北側約有{{space|1}}140{{space|1}}公尺長的路段，會短暫進入[[七股區]]後港里的範圍內。

在延平國小西北角路口南轉後，南27線回到[[佳里區]]境內時則已屬頂廍里的範圍，本線就以南偏東的方向到達頂廍里頂廓{{r|origin3}}及嘉福里下營{{r|origin4}}組成的聚落處；舊時這兩村落同屬下營庄，後才改劃分為兩個不同的村-{里}-{{r|sinicamaps}}。南27線在這兩村落間與[[南26線]]有一段短暫共線；共線結束後，本線轉向南行，沿下營東側的田間曲折道路，以大致東南的方向通往安西里大路尾{{r|origin5}}終點處，在此連接東西向的[[南32線]]，可經由此公路（文化路）往東進入[[佳里區]]主市區內。南27線沿線景觀就如同[[臺南市]]的農村道路相似，由農村、農地及小片林地間插組合而成，除了番子寮、頂廍、下營這幾處聚落比較熱鬧外，農田路段邊僅有幾處散戶居住。

南27線1976年以前的路線終點位於頂廍與下營間的''南28線''（此段路線後來被改編為現今的[[南26線]]）路口上。1976年改編後，併入「下營～大路尾」這段彎曲的田間路段，讓南27線可以由北到南橫向連絡[[南19線]]、[[南28線]]、[[南26線]]及[[南32線]]四條[[佳里區]]市區的連外道路，而成為現今所見的路線概況。
